Benzethonium Chloride or Hyamine or (Diisobutylphenoxyethoxyethyl)Dimethylbenzylammonium Chloride or Phemerol Chloride is a synthetic quaternary ammonium salt. Benzethonium chloride exhibits a broad spectrum of microbiocidal activity against bacteria, fungi, mold, and viruses. It is also found in cosmetics and toiletries such as soap, mouthwashes, anti-itch ointments, and antibacterial moist towelettes.

DEFINITION
Benzethonium Chloride contains NLT 97.0% and NMT 103.0% of benzethonium chloride (C27H42ClNO2), calculated on the dried basis.
IDENTIFICATION
A.
Sample solution: 10 mg/mL.
Analysis: Add 2 mL of alcohol, 0.5 mL of 2 N nitric acid, and 1 mL of silver nitrate TS to 1 mL of the Sample solution.
Acceptance criteria: A white precipitate, which is insoluble in 2 N nitric acid but soluble in 6 N ammonium hydroxide, is formed.
B. Infrared Absorption.
C. The retention time of the major peak of the Sample solution corresponds to that of the Standard solution, as obtained in the Assay.
Residue on Ignition: NMT 0.1%
Melting Range or Temperature: 158C –163C, the specimen having been dried previously.
Loss on Drying: NMT 5.0% at 105C for 4 h.

DEFINITION
N-Benzyl-N,N-dimethyl-2-[2-[4-(1,1,3,3-tetramethylbutyl)phenoxy]ethoxy]ethanaminium chloride.
Content: 97.0 per cent to 103.0 per cent (dried substance).
CHARACTERS
Appearance: White or yellowish-white powder.
Solubility: Very soluble in water and in ethanol (96 per cent), freely soluble in methylene chloride.
An aqueous solution froths copiously when shaken.
IDENTIFICATION
A. Melting point: 158C to 164C, after drying at 105C for 4 h.
B. Thin-layer chromatography.
C. To 5 mL of dilute sodium hydroxide solution add 0.1 mL of bromophenol blue solution and 5 mL of methylene chloride and shake. The lower layer is colourless. Add 0.1 mL of solution S and shake. A blue colour develops in the lower layer.
D. To 2 mL of solution S add 1 mL of dilute nitric acid. A white precipitate is formed which dissolves upon addition of 5 mL of ethanol (96 per cent). The solution gives reaction (a) of chlorides.
Appearance of solution: Solution S is clear and not more intensely coloured than reference solution.
Acidity or alkalinity: To 25 mL of solution S add 0.1 mL of phenolphthalein solution. The solution is colourless. Add 0.3 mL of 0.01 M sodium hydroxide. The solution is pink. Add 0.1 mL of methyl red solution and 0.5 mL of0.01 M hydrochloric acid. The solution is orange-red.
Volatile bases and salts of volatile bases: Maximum 50 ppm, determined on 0.20 g.
Loss on drying: Maximum 5.0 per cent, determined on 1.000 g by drying in an oven at 105C for 4 h.
Sulfated ash: Maximum 0.1 per cent, determined on 1.0& g.
